Inhibition of PLK1 rescues LKB1 deficiency-induced centrosome amplification. U2OS/shRNA-LKB1 and U2OS/shRNA-Ctrl cells were transfected with siRNA of PLK1 or scramble control for 48 h. The cells were then fixed for immunofluorescent detection of PLK1 and pericentrin (a and b). (a) Fluorescent images were captured with an Olympus IX53 microscope. (b) Percentage of cells with >2 centrosomes was calculated. The graph represents the mean and S.D. of three independent experiments where at least 50 cells were counted per experiment. *P<0.05. (c) Lysates of siRNA PLK1 or control-transfected cells were harvested and applied for immunoblot. For each lane, 30 μg proteins were loaded. GAPDH served as a loading control
