Subcellular localization of GmWRKY31 protein and transactivation assays of GmWRKY31 to the W-box element. (A) Subcellular localization was investigated in Arabidopsis mesophyll protoplasts under a confocal microscope. The fluorescent distribution of humanized hGFP, the fusion protein GmERF5-hGFP and GmWRKY31-hGFP were observed under white light, UV light, and red light, respectively. Bars, 10 μm. (B) Schematic diagram of effector, reporter and internal control plasmid constructs. Effector plasmids encoded GmWRKY31, under the control of the CaMV35S promoter. Reporter plasmids contained three repeats of the wild type or mutant W-box and 35Smini. (C) Relative GUS activities in transactivation assays. The effector plasmid encoding GmWRKY31 and the reporter plasmid were co-transfected into Arabidopsis mesophyll protoplasts with internal control plasmid by polyethylene glycol-mediated transformation. The numbers showed the fold increase in GUS activity compared with the vector GmWRKY31+35S mini. Results were averages of three replicates ± standard deviation. Statistically analyzed using Student’s t-test (P < 0.05, ∗∗P < 0.01).
