Kinetic plot representing the evolution of the total amount of free thiol following an exposure to 1 mM H2O2. The total fluorescence intensity measured for each lane at 572 nm was corrected by the total amount of protein in a given lane estimated by Coomassie Blue staining using ImageJ. The plot shows a rapid decrease of the amount of free thiol as expected due the presence of the oxidative reagent. The variation is quickly inversed after 15 min. The point at 30 min corresponding to the original OxiTMT experiment shows a higher amount of free thiol caused by the induction of anti-oxidant proteins as shown by proteomics results and appears as a plateau: this general reducing environment is maintained 60 min after the exposure to H2O2
