Figure 2.

Biochemical properties of TaRecJ1. A and B, TaRecJ1 (WT) and its mutant TaRecJ1 D41A/D43A (mt) were incubated at 50 °C with 50 nm 3′-FITC-labeled 54-nt ssDNA (HJ3–54-mer), 54 bp of dsDNA (HJ3–54-mer + HJ3–54-mer RC), 34 bp dsDNA with a 20-nt 5′-overhang (HJ3–54mer + trap DNA), or 34 bp of dsDNA with a 20-nt 3′-overhang (HJ3–54mer + HJ3RC34). The reaction products were separated by native 20% PAGE (A) and denaturing 15% PAGE (B). Asterisks indicate the positions of fluorescent labeling. C, quantification of the degraded products shown in A. The decreased amount of substrate DNA was quantified and plotted at each time. The averages in three independent experiments with the S.E. are shown.
