Effect of the TaGINS·TaRecJ2 complex on the helicase activity of TaMCM. A, the Cy5-labeled splayed-arm DNA substrate (50 nm) was incubated with TaMCM and increasing amounts of TaRecJ2. The reaction was performed at 60 °C for 15 min. B, the same substrate was incubated with 50 nm TaMCM, 500 nm TaGINS, and increasing amounts of TaRecJ2 (WT) or TaRecJ2-D34A/D36A (mt). The reaction was performed at 50 °C for 15 min. The helicase activity is expressed at the bottom of the panel as the relative amount of unwound DNA (%) in each reaction condition. ssDNA was loaded in parallel as controls for the unwinding reaction.
