Intracellular 2-OG concentrations of E. coli 20 mL of exponentially growing E. coli culture were rapidly cooled to 0 °C and harvested by centrifugation. Two E. coli cultures (A,B) were used and two samples were taken per culture (1, 2). 2-OG was extracted and intracellular concentrations determined by FRET-sensor (grey; error bars: ±s.e.m. with n = 4) or GC-MS measurements (white; error bar denotes the standard error of the 2-OG calibration curve).
