Expression from a high copy number plasmid results in higher amounts of crRNAs in the cell. RNA was isolated from cells expressing crRNA#3 from a high copy number plasmid (lane high) or a low copy number plasmid (lane low), separated by an 8% polyacrylamide gel electrophoresis (PAGE) and transferred to a membrane that was subsequently hybridized with a probe against the crRNA (upper panel crRNA) or a probe against the 5S rRNA (lower panel).
