Immunoblot assays showing the fluence-rate response of the blue light-dependent phosphorylation of CRY2 catalysed by PPK1 (a), PPK2 (b), PPK3 (c) and PPK4 (d) in HEK293T cells. Cells co-transfected to express CMV::Myc-CRY2 and indicated individual CMV::Flag-GFP-PPKs were exposed to the different fluence rates of blue light (0–100 μmol m−2 s−1) for 30 min. CRY2 and PPK were detected by immunoblots probed with anti-Myc or anti-Flag antibodies, respectively. Arrowheads and arrows indicate phosphorylated CRY2 (upshift bands) and unphosphorylated CRY2, respectively.
