The subcellular distribution of HIV-1SF2 Nef wild type and mutants is largely preserved in rodent and human cell lines. Human and rodent cells constitutively expressing human CD4 and CCR5 were plated on coverslips and transfected with expression constructs encoding the indicated Nef.GFP fusion proteins or GFP alone. One day after transfection, the cells were fixed with paraformaldehyde, and digital images of GFP fluorescence were acquired on an Olympus IX 70 microscope. Scale bar, 20 μm.
