Decreased Suf function results in increased sensitivity to RNS and ROS. (A) Methyl viologen sensitivity was monitored in the WT, sufD*, sufD* suf+, and sodA::Tn strains. The cells were cultured in TSB before serial dilution and spot plating on solid TSA supplemented with 40 mM methyl viologen or vehicle control. (B) H2O2 sensitivity was assessed in the WT, sufD*, sufD* suf+, and katA::Tn strains. The cells were challenged with 500 mM H2O2 before the reaction was quenched, and the cells were serially diluted and spot plated on solid TSA medium. (C) Nitroprusside (NP) sensitivity was assessed in the WT, sufD*, and sufD* suf+ strains. Representative growth profiles in the presence and absence of 15 mM nitroprusside in TSB medium are shown.
