Hop1CTD, but not full-length Hop1 or HORMA, exhibits a high affinity binding to the i-motif formed by meiosis-specific DSB from chromosome IV of S. cerevisiae. Reaction mixtures (20 μL) containing 0.5 nM 32P-labeled C-rich ssDNA (AC) or i-motif mutant DNA (DF) or i-motif DNA (GI) were incubated in the absence (lane 1) or the presence of (lanes 211) 150, 300, 450, 600, 750, 900, 1050, 1200, 1350, and 1500 nM of full-length Hop1, HORMA, or Hop1CTD, respectively. The solid triangle on top of the gel denotes increasing concentrations of the specified protein.
