(A) Conditioned medium (CM) from both unstimulated and LPS-pretreated macrophages (LPS-CM) stimulated miR-21 expression in vascular smooth muscle cells (VSMCs) and primary adventitial fibroblasts. (B) Screening with qPCR for potential macrophage-derived cytokines that could modulate miR-21 expression tested in NIH3T3 cells. (C) Quantification of the concentration of TNF-α and TGF-b in CM and LPS-CM using ELISA. (D) The stimulating effects of conditioned medium on miR-21 expression were blocked by a neutralizing antibody to TNF-α in primary fibroblasts and VSMCs.
