Tempol inhibits KO₂-induced TNFα, IL-1β, and IL-10 mRNA expression in the paw skin and spinal cord. Mice were treated with tempol (100 mg/kg) or vehicle 40 min before KO₂ intraplantar injection. Paw skin and spinal cord samples were collected 3 h after KO₂-stimulus and TNFα ((a) and (b)), IL-1β ((c) and (d)), and IL-10 ((e) and (f)) mRNA expressions were evaluated by RT-qPCR. Results were normalized using GAPDH as gene control and expressed as means ± SEM (n = 6 per group per experiment, representative of two separate experiments) [^∗p < 0.05 versus saline; ^#p < 0.05 versus KO₂ + vehicle (one-way ANOVA followed by Tukey's t-test)].
