Effect of pre-treatment of MK-8776 on radiation-induced double-strand breaks and cellular radiosensitivity. (A and B) After EMT6 cells were treated with vehicle (black) or MK-8776 (white) for 1 h, they were X-irradiated at 1 Gy and incubated for the indicated times in the presence or absence of drug. At least 100 cells were analyzed and the average number of 53BP1 (A) or γ-H2AX (B) foci per cell was scored. Data are expressed as means ± S.D. from three experiments. * P < .05, ** P < .01 vs. radiation alone (Student's t-test). (C) After EMT6 cells were treated with vehicle (black) or MK-8776 (white) for 1 h, they were X-irradiated and incubated with or without drug for 18 h. Subsequently, media was replaced and the cells were cultured for 7 days in the absence of drug. Cellular radiosensitivity was determined by clonogenic survival assay. The surviving fraction was calculated using a correction for plating efficiency without X-irradiation. Data are expressed as means ± S.D. from three experiments.
