Effects of an interleukin (IL)-25 pretreatment on synoviocyte IL-6 expression and production. (A) Cells were pretreated for 4 h with 50 ng/mL IL-25 and then treated with IL-17A (50 ng/mL) and/or tumor necrosis factor alpha (TNF-α) (0.5 ng/mL). RNA was extracted after 12 h and supernatants were collected after 24 h. The production of IL-6 was quantified by enzyme-linked immunosorbent assay (ELISA) and the expression of IL-6 was assessed by qRT-PCR. *p < 0.05 (Mann–Whitney non-parametric test). (B) Cells were transfected with 0.5 µg siRNAs targeting IL17RA, IL17RB, and IL17RC alone or in combination. After 48 h, RNA was extracted and the expression of IL17RA, IL17RB, and IL17RC was analyzed by qRT-PCR. **p < 0.01 and *p < 0.05 (Mann–Whitney non-parametric test).
