ISRE induction by plasmid DNA transfection is boosted in ChX710-treated cells. (a) ISRE-luciferase reporter cells were transfected with 75 ng/well of plasmid DNA (pCiNeo-3xFlag-GW), and cultured at 4 × 104 cells/well in 96-well plates with DMSO alone or increasing concentrations of ChX710. After 24 hours of incubation, luciferase induction was determined. (b) Same experiment as in (a) but cells were treated with 6 μM of ChX710 or CID70, an inactivate analog of ChX710. (c) Same experiment as in (a) but cells were transfected with 25 ng/well of pCiNeo-3xFlag-GW (Plasmid-1), pTK-Renilla (Plasmid-2) or pDONR207 (Plasmid-3), and stimulated with ChX710 at 6 μM. Experiments were performed in duplicate, and data represent means ± SD. ***P < 0.01 as calculated by two-way ANOVA and Bonferroni’s post hoc test (a and b) or Student’s t test (c).
