Rough optimal temperature determination for the trans-cleaving catalyst 925-11t. Cleavage reactions were performed under multiple turnover conditions with 100 nM catalyst, 30 μM unlabeled S1 and <1 nM 5′ 32P-labeled S1 in buffer. Values of kobs MTR were obtained from the slope values obtained from linear regression (Equation 2), divided by the catalyst concentration (Equation 3, [E]T = 100 nM) and plotted against temperature to give a temperature optimum near 24°C for the first-order rate constant for cleavage of S1 in S1 saturating conditions.
