Refinement of regions mediating WAVE2 binding to Abi-1. (A) Lysate proteins prepared from Cos-7 cells cotransfected with the Abi-1 cDNA and DsRed-WAVE2 full-length cDNA or DsRed-WAVE2 cDNAs with the indicated amino acid deletions (▵) were immunoprecipitated with anti-Abi-1 antibody and the precipitated proteins subjected to immunoblotting analysis with anti-DsRed and then anti-Abi-1 antibody (Abi-1 precipitates) or anti-DsRed antibody alone (lysate proteins). (B) Peptides (20 mers) from across the Abi-1 coiled-coil domain (amino acids 45–107) were synthesized and spotted on a cellulose filter, the filter then incubated with GST-WAVE2 WHD, and bound fusion detected by immunoblotting with anti-GST antibody.
