Abi-1 deficiency impairs WAVE2 membrane translocation and cell spreading and migration. (A) Lysate proteins from B16F1 cells expressing scrambled (pSiSc) or Abi-1 silencing (pSi-Abi-1) short-hairpin RNAs were immunoblotted with the indicated antibodies (Left) or plated on FN, stained for actin, and examined by immunofluorescent microscopy (Right). (B and C) pSiSc or pSi-Abi-1 B16F1 transfectants were plated on FN (C) or on FN in the presence of fluorescent latex beads (D), the cells fixed and stained after 18 h, and average cell areas and tracking distances evaluated by using an Arrayscan II Reader (Cellomics, Pittsburgh). Values represent means (±SEM) of two independent experiments. (D) B16F1 cells transfected with the pSiSc and pSi-Abi-1 vectors were plated on FN for 6 h, stained for actin, and Rac and WAVE2 and images examined by immunofluorescent microscopy.
