Assembly of RSV CA tubes followed using classical light scattering. (A) Dependence on Temperature. RSV CA (300 μM) in 12.5 mM MOPS/KOH pH 7.0, 75 mM NaCl, 10%(w/v) Ficoll 400 was incubated at varying temperature. (B) Dependence on Protein Concentration. RSV CA (at varying concentration) in 12.5 mM MOPS/KOH pH 7.0, 75 mM NaCl, 10%(w/v) Ficoll 400 was incubated at 34 °C. (C) Dependence on Neutral Salt Concentration. RSV CA (350 μM) in 12.5 mM MOPS/KOH pH 7.0, varying NaCl concentration, 10%(w/v) Ficoll 400 was incubated at 34 °C. Absorbance at 250 nm was followed as a function of time in each case. The displayed absorbance data are the mean of replicate measurements made under each condition.
