M. marinum ΔwhiB4 was less potent inducing TNF. RAW 264.7 macrophage cells were infected with mycobacteria at MOI of 10. At 3, 12, and 24-hr post infection, cell-free supernatants were collected and the levels of TNF were determined by enzyme-linked immunosorbent assay (ELISA). Data are plotted as mean ± SEM (n = 3), and are representative of two independent experiments. The level of TNF secretion induced by ΔwhiB4 was significantly lower than that of WT or the complemented strain (***p < 0.001; two-way ANOVA).
