Figure 4. Deguelin suppresses HK2 localization on mitochondria and induces apoptosis.

(A) deguelin inhibits HK2 localization on mitochondria and up-regulates cleaved-PARP. H460 (left), HCC827 (middle) and H1650 (right) cells were treated with deguelin for 24 h as indicated, the mitochondrial fractions and whole cell extracts were isolated, western blot was conducted to detect target proteins. (B) deguelin induces apoptosis. H460 cells were treated with deguelin at the indicated concentrations for 24 h, flow cytometry was conducted as described in the “Material and Methods” section. Each experiment was conducted 3 times independently and the data are shown as means ± S.D., the asterisks indicate a significant increase (***p < 0.001) of deguelin induced apoptosis.
