(A and B) deguelin disrupts the interaction of HK2 and VDAC1. H460 (A) and HCC827 (B) cells were treated with deguelin for 24 h, co-immunoprecipitation was conducted to detect the interaction between HK2 and VDAC1. (C and D) overexpression of HK2 rescues deguelin-induced mitochondrial apoptosis in H460 cells. The HK2 or vehicle plasmid was transfected into H460 and HCC827 cells, after 24 h, these cells were treated with deguelin for another 24 h as indicated. Western blotting was performed to detect the protein expression in whole cell extraction (C) and subcellular fractions (D).
