Effect of down-mutations in the ribosome-binding site on the response to phosphosugar stress of the cytoplasmic ptsG-crp mRNA. (A) IT1568 harboring pMM12FS (lanes 1,2) or pMM12FSM (lanes 3,4) was grown with 1% Glc or αMG to A₆₀₀ = 0.6. Total proteins were prepared, and the indicated amounts of samples were subjected to Western blot analysis using anti-CRP antibody. (B) Cells were grown to A₆₀₀ = 0.5. Glc or αMG was added at a final concentration of 1%, and incubation was continued for 20 min. Total RNAs were prepared, and the indicated amounts of RNA samples were subjected to Northern blot analysis using the crp probe.
