Heterokaryon assays. Semiconfluent monkey Vero cell monolayers were transfected with the pGFP-p17 (A) or pGFP-p17(104-146) (B) plasmid by the use of Lipofectamine, and 24 h later the cells were scraped off the plate. Aliquots of 5 × 10⁵ transfected cells were then fused with 5 × 10⁵ untransfected mouse L cells in the presence of 50% polyethylene glycol 4000 and 100 μg of cycloheximide/ml. At the indicated postfusion times, the cells were fixed and stained with Hoechst 33258.
