Glucocorticoids mediate MAPK signaling pathway by TGF-β1 in A549 cells. Cells were pretreated with or without dexamethasone (Dex, 2.5 μM) and fluticasone propionate (FP, 2.5 μM) or the MAPK pathway inhibitors of p38 (SB203580, 10 μM) or ERK (U0126, 10 μM) and then stimulated with TGF-β1 (5 ng/ml) for 30 minutes. (A) Phosphorylation of MAPK signaling molecules: p-p38, p-ERK and total MAPK signaling molecules were determined by Western blot. Data were normalized to total p38 or ERK expression. (B) Protein levels of EMT-related markers were determined by Western blot. Data were normalized to β-actin. Data are presented as mean ± SEM. Results were from at least three independent experiments. *p < 0.05 vs. control; †p < 0.05 vs. TGF-β1.
