2Apro and 3Cpro proteases did not cleave KPNA1 or inhibit IFN-β response. A and B, HEK293 cells were transfected with pRK5-Flag-3C (A) or pRK-HA-2A (B), respectively, and the cell lysates were prepared for Western blot analyses with antibodies specific for KPNA1, TRIF, or eIF4G. C, HEK293 cells were co-transfected with ISRE reporter plasmids, along with either pRK5-HA-2A, pRK5-Flag-3C, or a control plasmid at the indicated doses, respectively. At 24 h post-transfection, the cells were stimulated with 20 ng/ml IFN-β. The cell lysates were collected 16 h later and analyzed for luciferase activities.
