HEK293 cells were transiently transfected with an expression vector for EGFP and either human wild-type TRPM2 or a channel carrying the indicated mutation. At 24 h post transfection, cells were loaded with Fura2 and subjected to calcium imaging. At the indicated time, either buffer or hydrogen peroxide (final concentration 1 mM) was added. Analysis was restricted to EGFP-positive cells. For differentiation between responding and non-responding cells, the indicated threshold of 0.7 was chosen.
