a 864669, b 4248543, and c 856350 downmodulates IL6-induced upregulation of pSTAT3Tyr705 and decreases STAT3 target gene expression. Cal33 cells were serum starved and treated with increasing concentrations of 864669, 4248543, and 856350, respectively. At the end of 2 h and 45 min, cells were stimulated with IL6 (50 ng/ml, 15 min) and harvested and protein lysates were prepared and ran on a 10% SDS-polyacrylamide gel. A significant decrease in pSTAT3Tyr705, cyclin D1, and Bcl-XL expression was observed in Cal33 cells treated with 864669 (Fig. 2a) and 4248543 (Fig. 2b), and 856350 (Fig. 2c) at 30 μM compared to cells stimulated with IL-6. The only exception was Bcl-XL expression (Fig. 2c) in Cal33 cells treated with 856350 where a dose-dependent decrease was observed although it was not significant.
