Target RNA structure can inhibit siRNA binding. (A) Radioactively labeled oligonucleotide simulating the antisense strand of siRNA-Nef was incubated with an increasing amount of wild-type or R8 RNA. The siRNA/target-RNA duplex formation was analyzed by EMSA. (B) Free and bound siRNA oligonucleotide was quantified to calculate the level of duplex formation (bound siRNA/free + bound siRNA).
