Single-cycle HIV-1 transmission assay. Immature DC were stimulated for 48 h with poly(I·C); gamma interferon (IFNγ) plus LPS, IL-1β, and TNF-α; and PgE2 plus LPS, IL-1β, and TNF-α to produce two types of DC1 and one type of DC2, respectively. The DC were incubated for 2 h with different amounts of HIV-1 (0.3, 3, and 30 ng of CA-p24), followed by washing steps to remove unbound virus.
