Relative 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) content in pronuclear stage embryos. Nuclear DNA was extracted from pronuclear stage embryos in the absence or presence of 500 ng/mL ascorbic acid. 5-mC content was analyzed by real-time polymerase chain reaction. Ctrl, control group; VC500, embryos treated with 500 ng/mL ascorbic acid; TET3KD, TET3 knock down; TET3 KD+VC, TET3 was knocked down and embryos were treated with 500 ng/mL ascorbic acid. Values are mean±standard error of the mean of three independent experiments. Asterisks (*) indicate p<0.05.
