Andrographolide analogs inhibited pro-inflammatory cytokines expression. THP-1 cells differentiated with 10−8 M PMA were treated with 1 μg/ml LPS in the presence or absence of 3b and 5a for 6 h. The levels of secreted (A) IL-1β, (B) TNF-α, and (C) IL-6 in the medium was determined by LEGENDplexTM bead-based immunoassays. (D) THP-1 cells were transfected with 50 ng/μl poly I:C in the presence or absence of 3b and 5a for 9 h. The level of secreted TNF-α in the medium was determined as in (B). Effects of 3a, 5b, and 6b on the LPS-stimulated secretion of (E) IL-1β, (F) TNF-α, and (G) IL-6 were determined as in (A).
