Figure 11.

JA Hypersensitivity of 35S:MYC2-GFP (MYC2OE) Plants Can Be Further Enhanced by Overexpression of UBP12 and UBP13.

(A) Seedling phenotypes with MeJA treatments. Seedlings of wild type (Col-0), XVE:UBP12-HA/MYC2OE, XVE:UBP13-HA/MYC2OE, 35S:MYC2-GFP (MYC2OE), and jin1-9 were grown vertically on MS medium containing with or without MeJA (5 μM) and with and without β-estradiol (10 μM) for 14 d. Bars = 10 mm.

(B) Quantitative evaluation of primary root length of seedlings in (A). The y axis shows average values of primary root length. Bars represent average values ± sd of more than 25 seedlings of each genotype. The average primary root length of each seedling group on MS was fixed as 100%, and the relative primary root length of each genotype with MeJA is represented as the percentage of that without MeJA. **P < 0.01 (LSD); one-way ANOVA. Results of statistical analyses are presented in Supplemental File 1.

(C) Quantitative PCR analysis of expression of JA response genes and senescence-related genes. RNA samples were prepared from seedlings in (A). ACT2 expression levels were used as an internal control. Bars represent average value ± sd (n = 3 independent pools of 25 seedlings per pool). **P < 0.01 and *P < 0.05; two-tailed t test.
