Figure 7.
Plants of UBQ10:UBP12-HA and UBQ10:UBP13-HA Are Hypersensitive to MeJA.
(A) Phenotype of seedlings with MeJA treatments. Seedlings of wild type (Col-0), UBQ10:UBP12-HA, UBQ10:UBP13-HA, 35S:MYC2-GFP (MYC2OE), and jin1-9 were grown vertically on MS medium with or without 10 μM MeJA for 14 d. Bars = 10 mm.
(B) Quantitative evaluation of primary root length of seedlings in (A). The y axis shows average values of primary root length. Bars represent average values ± sd of more than 25 seedlings for each genotype. The average primary root length of each genotype group on MS was fixed as 100%, and the relative primary root length of each genotype with MeJA is represented relative to that without MeJA. **P < 0.01 (LSD); one-way ANOVA. Results of statistical analyses are presented in Supplemental File 1.
(C) Protein levels of UBP12 and UBP13. Proteins were extracted from 10-d-old wild type (Col-0), UBQ10:UBP12-HA, and UBQ10:UBP13-HA seedlings and analyzed by immunoblots. UBP12-HA and UBP13-HA were detected by anti-HA antibody. β-Tubulin levels were used as a loading control.
(D) Quantitative PCR analysis of expression of JA response genes and senescence-related genes. RNA samples were prepared from seedlings in (A). ACT2 expression levels were used as an internal control. Bars are average values ± sd (n = 3 independent pools of 25 seedlings per pool). **P < 0.01 and *P < 0.05; two-tailed t test.
