(A) THP-1 differentiated macrophages were transfected with NF-κB, STAT3 and AP-1 luciferase reporter constructs and then treated with indicated concentration of Btk inhibitor in the presence or absence of LPS stimulation (1 μg/ml) for 6 hours prior to the measurement of luciferase. (B) THP-1 differentiated macrophages were pretreated with the indicated concentrations of Btk inhibitors for 1 hour and then stimulated or not with LPS (1 μg/ml) for 10 minutes. The nuclear extracts of cultured cells were prepared and probed by Western blot analysis to measure p-p65, p-STAT3 and p-AP1 levels. PCNA is shown as a loading control. *Significantly decreased compared to LPS treatment alone (p < 0.05). Results are representative of three similar experiments.
