α-Toxin preferentially eradicates mature erythroblasts. (A,B) A total of 5 × 106 bone marrow cells were cultured for 24 hours in the presence of 10 ng/ml α-toxin (Wild-type) or a variant α-toxin (H148G), and flow cytometry analysis was performed using a Guava easyCyte. (A) A representative flow cytometry profile is shown. (B) The absolute number of CD71+TER119, CD71+TER119+, and CD71TER119+ cells per culture well was determined. (C,D) Bone marrow cells were treated with the indicated concentration of α-toxin (Wild-type) or a variant α-toxin (H148G), and the cells were plated in Methocult 3334.
