F-actin is required for the biogenesis of secretory granules. COS7-CgA cells (a) and PC12 cells (b) were treated using DMEM alone (control) or supplemented with latrunculin B. They were labeled with fluorescent phalloidin (F-actin), immunolabeled with anti-CgA antibody and analyzed by confocal microscopy. Projected images of 26 0.3 µm-thick confocal sections throughout the cells are shown. The scale bar represents 20 µm. The number of CgA granules in control and treated cells was quantified and expressed as mean + s.e.m. from three independent experiments (n = 30 cells). *P < 0.05, ****P < 0.0001 (Mann-Whitney test).
