UTP-Ca2+ responses depend on the phospholipase C (PLC) signaling pathway in human BON. The PLC inhibitor U73122 (10 μM) abolished the Ca2+ responses induced by UTP (A) in cells with Ca2+ oscillations or (B) monophasic Ca2+ transients, (C) and reduced Ca2+ responses in other cells. Representative Ca2+ responses are shown in (A–C) and histograms associated with each type of response represent pooled data showing that U73122 can abolish or reduce Ca2+ responses. (D–F) Average population response showing that U73122 but not its inactive analog U73343, can block UTP-Ca2+ responses in BON (EC) cells. Cells were pre-incubated with U73122 or U73343 for 10 min.
