Gingipain deficiency reverses the suppression of F. nucleatum invasion by P. gingivalis. HOK-16B cells were infected with CFSE-labeled F. nucleatum and unlabeled wild-type P. gingivalis or gingipain mutants at a MOI of 500 for 4 h. (A) Cells were analyzed using flow cytometry after quenching the extracellular fluorescence from the bacteria attached to the cell surface with trypan blue. The percentage of cells containing F. nucleatum (left panel) and the MFI (right panel) are shown as the mean ± standard deviation. Representative data from three independent experiments are shown. (B) The cells were examined under a confocal laser scanning microscope after staining for F-actin (red) and nuclei (blue). CFSE-labeled F. nucleatum is displayed in green.
