The role of JDA-202 in cell cycle arrest in EC109 and KYSE-450 cells. EC109 and KYSE-450 cells were treated with JDA-202 at the indicated concentrations (0, 10, 20, and 40 μM). After treatment for 12 and 24 h, cells were washed with PBS and fixed in 70% ethanol overnight at 4°C. The cells were then stained by PI and analyzed by flow cytometry in EC109 (A, B) and KYSE-450 (E, F) cells; (C, D) and (G, H) are the quantitation of those cell cycle distributions. EC109 and KYSE-450 cells were treated by JDA-202 (0, 10, 20, and 40 μM) for 24 h; the expression levels (I, K) and quantitation (J, L) of p53/phospho-p53 (Ser15), Cdc2/phospho-Cdc2 (Tyr15), and Cdk2/phospho-Cdk2 (Thr160) were detected by Western blot and Image J. Data are presented as means ± SD. Three individual experiments were performed for each group. *p < 0.05, **p < 0.01 as compared with the controls. PBS, phosphate-buffered saline.
