Growth phenotype and physiological responses of WT and PgTIP1-transgenic soybean lines under NaCl treatment. (A) The seedlings (T3 generation) of PgTIP1 transgenic soybean lines N and J11, and WT (cv. Nannong 8831) were screened by PCR. The PCR-verified seeds and WT were surface-sterilized, soaked, germinated, and planted in plastic pots containing quartz sand, fertigated with ½ Hoagland solution, and maintained in a greenhouse with a temperature ranging from 18 ± 2°C to 25 ± 2°C and a 14 h/10 h (day/night) photoperiod. At the time of the 1st trifoliate leaf expansion, the plants were subjected to salt stress by fertigating with ½ Hoagland solution plus 150 mM NaCl, or ½ Hoagland solution devoid of NaCl as the control treatment. The Hoagland solution treatments were replaced every 2 days. After 7 days treatment, (B) the growth phenotype was photographed, and measured (C) RWC in the 1st pair of unifoliate leaves, (D) root vigor, and (E) REL in roots and the 1st pair of unifoliate leaves. Each bar represents the mean and SD of three replicates, means in bars followed by different letters show significant difference (P < 0.05) between the parameters of WT and PgTIP1-transgenic lines.
