Table 1.

Effects of the fla3 mutation on flagellar length

Strain Average length (μm) Average length excluding zero length flagella (μm) % Aflagellate cells
Vegetative cells
   Wild-type (137c) 11.1 ± 3.5 (105) 11.6 ± 2.6 (100) 4.8
   fla3 8.0 ± 4.2 (107) 8.9 ± 3.5 (97) 9.3
   fla3::KAP-GFP 10.7 ± 3.1 (102) 11.1 ± 2.3 (98) 3.9
Gametic cells
   Wild-type (137c) 12.6 ± 3.8 (107) 13.2 ± 2.5 (102) 4.7
   fla3 9.4 ± 3.7 (85) 10.6 ± 1.3 (75) 11.8

Flagellar length measurements are expressed as mean ± SD (sample size). Cells were grown with aeration in TAP medium (vegetative growth) or transferred to M-N/5 for 4 h to induce gametogenesis. All measurements were made at 21°C. The mean flagellar lengths of fla3 cells are significantly shorter (p < 0.001) than those of wild-type cells for both vegetative and gametic cells. Rescue of fla3 cells with the epitope-tagged KAP restores flagellar lengths to wild-type levels.
