Coupling RNA pol II staining with nuclear DAPI staining shows that T. castaneum zygotic transcription starts between 3–6 hours after egg laying, when the energids reaches the periphery (Fig 2A-C-DAPI and E-G-RNA pol II). Similar results were obtained by western-blots using the same antibody (D).
