Streptococcal NADase regulates inflammasome-dependent release of IL-1β. (A) BMDMs of the indicated genotypes were primed with LPS and infected with wt, nga(G330D), or Δslo streptococci. ui, uninfected. (B) Differentiated and LPS-primed THP-1 cells were infected with wt, nga(G330D), or Δslo bacteria. (C to G) LPS-primed BMDMs were infected with wt, nga(G330D), Δslo, or ΔspeB GAS. Supernatants were analyzed, and secretion of IL-1β (A, B, C, and F) or IL-1α (D) was determined by ELISA or cytometric bead array (CBA), respectively. (E and G) Cell death was evaluated by measuring LDH release. Values were normalized to LDH release induced by wt bacterial infection of B6 macrophages. Graphs show means plus standard deviations (SD) (error bars) for triplicate samples and are representative of at least three independent experiments.
