nmMLCK mediates IL-1β induced claudin-3 downregulation in Caco2 cells. (A) Caco2 cells were treated with 100ng/nl IL-1β for 0, 6, or 24 hours, or (B) with 0, 10, or 100 ng/mL for 24 hours. Cell lysates were analyzed by Western blot and degree of claudin-3 expression was assessed by densitometry. β-actin was probed as loading control. (C) Caco2 cells were treated with vehicle control or ML-7 (10 μM) followed by IL-1β treatment (10 ng/mL or 100 ng/mL) and claudin-3 protein expression was assessed by Western blot. Results are representative of 3 experiments.
