Effects of W2476 on insulin and MafA expression. INS-1E cells were exposed to 5 mmol/L or 25 mmol/L glucose with or without varying concentrations of W2476 for 24 h, followed by measurement of mRNA levels of txnip (A), mafa (B) and insulin (C) using quantitative real-time PCR. Protein levels of TXNIP (D), MafA (D, E) and insulin (D, F) were evaluated by Western blotting. Representative immunoblots were shown and protein contents were separately quantified. Isolated rat islets of Langerhans were incubated for 48 h with 33.3 mmol/L glucose and 15 μmol/L W2476, and mRNA levels of txnip (G), insulin(H) and mafa (I) were assessed by quantitative real-time PCR.
