miR-29a decreased 5-hmC levels by directly targeting TET methylcytosine dioxygenases. (a) miR-29a target sites predicted by TargetScan for human, mouse, and zebrafish TET-family members. (b) miR-29a significantly suppressed the luciferase activity of TET family containing a wild-type 3'-UTR, but showed no effect on the activity of TET family with a mutant 3'-UTR. (c and d) Following transfection with miR-29a vector, qRT-PCR and western blotting experiments showed the mRNA and protein levels of TET1, 2, and 3 in SMMC-7721 and HepG2 cells were significantly decreased compared with controls.
