NEDD8 induces the sensitivity of cisplatin and radiation in NPC cells. (a,b) S18 cells with silenced NEDD8 and S26 cells with overexpressed NEDD8 were seeded in 96-well plates at a density of 1 × 103 per well and treated different concentrations cisplatin (CDDP) as indicated for 72 h. Cell viabilities were tested by the MTT assay. (c,d) The cells were seeded in 6-well plates at a density of 1 × 105 per well and treated with 8 μM CDDP for 48 h. The apoptosis rates were detected with the Annexin-V/PI kit and analyzed by flow cytometry. (e,f) The cells were treated with different GY of radiation. Representative images (e) and the inhibition rate of colonies formed (f) are shown. (g) Western blotting analysis of cleaved PARP, cleaved-caspase-3 and p53.
