ICG-001 attenuates the protective effects of LiCl on the osteogenic activity of MC3T3-E1 cells stimulated with Ti particles. MC3T3-E1 cells were pretreated with 10 μm ICG-001 for 30 min; then, 10 mM LiCl and 0.1 mg/ml Ti particles were added and cultured in osteogenic medium for the indicated times. (a and b) Cytoplasmic and nuclear extracts were isolated and analyzed by western blot analysis using β-catenin antibody. (c) ctnnb1 and (d) axin-2 mRNA levels were identified using RT-PCR after 24 h of incubation. (e) ALP activity, and (f) runx2 and (g) sp7 mRNA levels were determined after 3 days of incubation.
